Thrombophilia Profile - 4G/4G Polymorphism In The PAI-1 Gene

When you were diagnosed with a myeloproliferative neoplasm, did your hematologist/oncologist recommend a thrombophilia profile (pathogenic mutations for factor II, factor V, factor XIII, MTHFR, PAI-1 4G/4G or 4G/5G, etc.)
I found out that I have PAI-1 (Plasminogen-Activator Inhibitor-1) 4G/4G mutation, which means my body makes too much of an inhibitor of fibrinolysis, fibrinolysis, in simple terms, being the process that does not allow blood clots to grow or accumulate. If one has this… read more

I also was only tested for JKAK2 mutation. That's a good idea to have further genetic tests as it will guide current and future therapies.

I actually don’t know. I was tested for mutations and told I had the JAK2 positive ET. No technical info was given to me 😊

I have had the von Willebrand factors checked (due to the MPN) as well as MTHFR (due to another condition). My von Willebrand factors do drop below reference range when my PLT are high.

It sounds like having the PAI-1 4G/4G mutation is something to be aware of. I would think it would bear directly on your treatment decision for managing the MPN.

Tatiana, right or wrong, now take Asa 81 mg and variable Coumadin (warfarin) for APL.

Hi Tia or Tta, I have polycythemia-MF. Had a CVA , due to clotting disorder called anti phospholipid syndrome. Think this was related to PV-MF. I’m 73. My basic blood factors were ok. Jak2 positive. Was on Eliquis during CVA, but not Asa at that time.
